However, when you open the bag, you \u200cdiscover that it&#8217;s \u200bnot\u200d just coffee grounds; \u2063it also contains a mixture of sand and other foreign particles. This unexpected impurity completely ruins your experience.<\/p>\n<p>Just \u200bas you rely on the \u200bpurity of your coffee grounds \u200dfor a great cup of coffee, the pharmaceutical industry, \u200dincluding vaccine\u200b production, has regulations in place to ensure good manufacturing practices. Patients and consumers expect that\u2064 these guidelines\u2063 mean drug or \u200dvaccine formulations\u200b are \u200dfree from\u2062 unwanted substances, ensuring their safety and effectiveness.<\/p>\n<div class=\"my-5\">\n<p>Controlling impurities in\u2062 traditional chemical products is a \u200bwell-established practice, but for \u2064biological products like mRNA-based vaccines, managing impurities presents unique challenges.<\/p>\n<h2>mRNA Products Contain &#8216;Gene Factories&#8217;<\/h2>\n<p>Double-stranded\u200d DNA (dsDNA) is used to make the mRNA contained in the COVID-19 vaccines. Tiny dsDNA \u2064plasmids are\u200b small engineered gene factories (Figure 1). These factories produce the \u2064mRNA strands contained in\u200c the LNPs. A plasmid\u200d appears like a tiny micro-bracelet \u2064with different segments representing different pieces of \u2063genes.<\/p>\n<\/p><\/div>\n<div class=\"post-figure\">\n<figure style=\"width:600px\" class=\"aligncenter\"><figcaption>Figure 1. (The \u2063Epoch Times)<\/figcaption><\/figure>\n<\/p><\/div>\n<div class=\"my-5\">\n<p><b>Regulatory agencies like EMA\u2014Europe&#8217;s drug\u2064 regulatory \u2062authority\u2014set limits for the number of plasmids in the final \u200clots \u2064distributed for injection.<\/b> \u200c New \u2064questions \u2064have \u2062been raised \u200cabout\u200d how much contamination \u200cthere is, \u2062and whether\u2062 the FDA\u200b is monitoring \u2063this. It is\u200c also unclear whether the plasmids can merge\u200d with human genes\u200c within the cell \u200dor travel to the gut.<\/p>\n<p>The EMA standard for DNA contamination \u2063of vials is <a href=\"https:\/\/doi.org\/10.31219\/osf.io\/b9t7m\" target=\"_blank\" rel=\"noopener\">0.33 percent (330 \u200cpg\/mg)<\/a>, or roughly one DNA molecule for every 3,000 \u200dmRNA molecules. \u2063Although the Moderna \u200bmRNA-1273\u2062 vaccine \u2063meets this standard, \u2063the \u200cactual volume may be higher \u2062due to\u2064 poor quality control. The \u200dDNA must \u2064be\u200b removed \u2062from the final product before distribution, but some residual amount\u200d is expected to remain. Unanswered questions include: How much DNA\u2063 is in the vials? \u200bIs it over the\u200c limit? Is \u200bthe FDA tracking this? And what are the implications, \u200dif any, with \u200bregard to persistence in\u2064 the recipient?<\/p>\n<\/p><\/div>\n<p>There are \u2063at least two independent groups \u200dof investigators who have conducted lab tests and confirmed the mRNA vaccine of Pfizer has \u2062been contaminated by DNA.<\/p>\n<div class=\"my-5\">\n<p>One team of\u200b scientists, led by microbiologist \u2062Kevin McKernan,\u2064 <a href=\"https:\/\/doi.org\/10.31219\/osf.io\/b9t7m\" target=\"_blank\" rel=\"noopener\">published a preprint paper<\/a> showing that the \u2064Pfizer\/BioNTech\u200d BNT162b2 vaccine has \u2062DNA \u201corders of\u200d magnitude\u2064 higher\u2063 than the EMA\u2019s limit.\u201d His paper has not yet been peer-reviewed.\u2063 The batches examined, provided by an anonymous source, were unopened, expired vials that \u2064were not \u2063delivered on dry ice.\u2064 If these data hold, the actual\u2062 amount of plasmids was 18\u2063 to 70 times higher \u2063than the EMA limit. (Table\u200d 3, page 12.)<\/p>\n<\/p><\/div>\n<p>Obviously, future investigations should attempt to establish contamination\u200c levels using unexpired doses with an\u200b intact cold chain.<\/p>\n<p>Professor Phillip\u200c Buckhaults, who\u2064 holds a doctorate in biochemistry \u200cand molecular biology and is considered \u2064an expert in cancer genomics research at\u2064 the University of South Carolina, \u200bperformed\u200b an independent analysis for the presence\u2062 of DNA in Pfizer batches.<\/p>\n<div class=\"my-5\">\n    <a href=\"https:\/\/twitter.com\/RefugeOfSinner5\/status\/1703584056804548821\" target=\"_blank\" rel=\"noopener\">Professor \u2062Buckhaults<\/a> in testimony stated the following:\n  <\/div>\n<p>&#8220;The \u2064Pfizer vaccine is contaminated with plasmid \u200bDNA. It&#8217;s not just mRNA. It&#8217;s got bits of DNA in it. This DNA is the\u200c DNA vector that was\u2062 used\u2064 as the template for the in vitro transcription reaction when they made the mRNA. I know this is true because I sequenced \u2063it in my own lab.&#8221;<\/p>\n<div class=\"my-5\">\n<p>We will continue\u2062 to follow this line of research.<\/p>\n<h2>Theoretical \u2062Risk of Plasmid DNA Contamination<\/h2>\n<p>While having some DNA in a sample is unavoidable and deemed acceptable,\u2063 some\u200d have raised questions about the possibility for \u200cgenomic integration of the \u2062DNA. Our cells use\u2063 DNA in the nucleus\u2063 to \u200cmake protein, so if the plasmid \u200dDNA \u2062gets into the nucleus,\u2064 there is a \u2063theoretical risk that it can get transcribed and make a protein.<\/p>\n<p>About\u200b <a href=\"https:\/\/www.nature.com\/articles\/s41467-019-08333-8\" target=\"_blank\" rel=\"noopener\">5 to 10 percent of our \u2062human genome<\/a> contains\u2062 ancient retroviral DNA. However, this DNA is mutated \u2062to a\u2062 point that is\u2064 no longer harmful. Any further research on this\u200b topic will therefore need to establish not just the presence of DNA\u200b plasmid integration, but also its biological activity.<\/p>\n<\/p><\/div>\n<p>Professor Buckhaults \u2064further commented in his testimony:<\/p>\n<div class=\"my-5\">\n<p>&#8220;I am kind of\u2062 alarmed about the\u200d possible\u2063 consequences\u2062 of this\u2014both in terms of human health and biology\u2014but you\u200c should be alarmed about the\u200b regulatory process \u2063that\u200c allowed it to get there.&#8221;<\/p>\n<h2>Concerns About DNA Migration to the Gut<\/h2>\n<p>Related \u200bto\u200c the \u200dDNA contamination is the concern\u2063 about residual expression vectors, or plasmids, \u200bin the vials. To\u200b make \u2063a billion doses of mRNA vaccine, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7564888\/\" target=\"_blank\" rel=\"noopener\">more than a kilogram of DNA<\/a> is required. Plasmids help produce \u2063the DNA by splicing in the desired sequence into a bacterial\u200d plasmid (Figure 1).<\/p>\n<p>Then \u2064workhorse bacteria, often E. coli, help spin \u200dout the DNA for production. These bacteria have an extra burden: They must replicate not only their own genome but also the plasmid DNA inserted within their genome.\u2063 This takes\u2062 slightly more time,\u2064 so the bacteria without the additional \u2062DNA will eventually\u200c outcompete those with the DNA.<\/p>\n<p>To \u2064solve this problem,\u200c scientists also splice in an <a href=\"https:\/\/www.addgene.org\/mol-bio-reference\/#introduction\" target=\"_blank\" rel=\"noopener\">antibiotic resistance gene.<\/a> The \u200dentire pool of bacteria is then \u2062treated with an \u2062antibiotic to kill \u2064the \u2064faster-replicating\u2064 bacteria\u200d without the conferred \u200cantibiotic resistance. This selective elimination\u2062 allows the plasmid-carrying antibiotic-resistant bacteria to continue growing. In\u2062 other \u2062words,\u2063 this antibiotic \u2064resistance \u200bgene confers an advantage that drives selection pressure to \u2062favor the bacteria \u2062producing the desired DNA.<\/p>\n<p>However, some scientists are \u200cconcerned that exceeding EMA standards for DNA plasmid contamination could affect an already growing antibiotic resistance problem. This would be a potential concern\u200d only if plasmids containing the\u200c antibiotic resistance\u200c gene migrate to the gut, integrate with bacterial targets in the\u200d gut \u200dflora, and disrupt the microbiome of \u200dthe gut accordingly. Diseases, including \u2064obesity, diabetes, cardiovascular disorders, cancer, hypertension, \u200cand irritable bowel syndrome <a href=\"https:\/\/www.frontiersin.org\/articles\/10.3389\/fmicb.2022.999001\/full#:~:text=Several%20major%20human%20diseases%20are,individually%20later%20in%20this%20review.\" target=\"_blank\" rel=\"noopener\">have been \u200cloosely linked<\/a> with disturbances of the gut microbiome.<\/p>\n<h2>Truncated mRNA \u2063Contamination<\/h2>\n<p>Nucleic acid contamination with truncated, or shortened, mRNA fragments is something that EMA\u200b has \u2064been following since February 2021. On page 35 of the\u2062 EMA assessment\u2063 report\u2064 (<a href=\"https:\/\/www.ema.europa.eu\/en\/documents\/assessment-report\/comirnaty-epar-public-assessment-report_en.pdf\" target=\"_blank\" rel=\"noopener\">pdf<\/a>) on the BNT162b2 mRNA vaccine reviewed in Part 1,\u200c the EMA states, &#8220;Truncated and modified RNA are present as impurities.&#8221; The agency noted\u200b that \u2064the impurities were \u2064found at different levels during \u2063production. \u200dFor instance, levels \u200cmay be higher in smaller test batches than in larger\u200d commercial batches.<\/p>\n<p>In fact,\u200d Danish scientists, Max Schmeling, Vibeke Manniche, and Peter\u200b Riis Hansen<a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/eci.13998\" target=\"_blank\" rel=\"noopener\"> linked adverse events with vaccination records<\/a> and found that smaller batches \u200bof the BNT162b2 \u200bmRNA vaccine\u200d may have a higher rate of adverse events (AEs). While this\u2064 finding\u200b is intriguing, the \u2064authors call\u200c for\u200b more \u2062research to see if this\u200d is a consistent pattern. We reviewed\u2062 the raw \u2062data\u2064 provided\u2063 by the authors and agree that a clustering of AEs seems to be found with batches having fewer than 100,000 doses.<\/p>\n<p>It has already been demonstrated in\u2064 vitro in a\u2064 laboratory experiment that mRNA can be<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/35723296\/\" target=\"_blank\" rel=\"noopener\"> reverse-transcribed to DNA<\/a> within six hours. A remaining question \u200bis\u2062 whether this\u2063 can\u200b happen\u200d in a live\u200b organism.\u200b Thus far, there is no evidence that\u2064 a reverse-transcribed DNA product can merge \u2062with a human cell\u2019s genome. Claims about integration are solely speculative\u200d and based upon an evolutionary precedent \u2063for such a process.<\/p>\n<p>The \u2064EMA asked for additional testing but allowed distribution to go forward. The scientists \u200dbelieved these fragments were unlikely\u200c to be\u2062 intact mRNA fragments. An intact mRNA fragment needs to have a cap \u200dand \u200da tail. The cap \u2064and tail are needed to tell\u200c the cell \u200bwhen to start \u200band stop producing the spike protein.<\/p>\n<p>Nevertheless, \u2063the EMA requested additional reports. The agency \u200cwas concerned that\u200d an autoimmune reaction could be triggered if\u200c fragments&#8217; potentially \u2064encoded\u200b proteins resemble\u2064 human proteins. In other words, if the\u200b fragments \u200d&#8221;mimic&#8221; human \u200bproteins, antibodies could \u200bbe developed against our \u2062own bodies.<\/p>\n<p>&#8220;Any homology between translated proteins (other than the intended spike protein) and \u2063human\u2064 proteins that\u200d may, \u200ddue to\u2064 molecular mimicry,\u200d potentially \u200dcause an autoimmune process should be evaluated. Due date:\u200b July 2021. Interim reports: March 2021, and on a monthly basis,&#8221; the\u200c EMA stated.<\/p>\n<p>It is clear that the mass production of mRNA at an industrial scale \u2062carries potential risks. This issue has\u2062 been raised recently by other researchers pointing out the \u2062role of<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S0264410X21012809?pes=vor#b0100\" target=\"_blank\" rel=\"noopener\"> manufacturing quality control<\/a>.
