This gene plays a role in regulating cell function \u2063and transmitting signals. <\/p>\n<p>The mutation in LRRK2 leads\u200b to an \u200boverproduction of a protein called \u200cdardarin, rather \u200bthan causing it to \u200cbecome deformed. Until now, scientists were unsure of how to control this protein expression due to a lack of understanding of the underlying mechanisms.<\/p>\n<p>In a recent publication in The \u2062Embo Journal, Xiong \u200drevealed that her lab team has solved this mystery by identifying an enzyme called ATIC, which acts \u200das a regulator for LRRK2, and a potential pharmaceutical \u2063treatment. <\/p>\n<p>To find potential candidates for genes that\u2063 control LRRK2, the researchers conducted a genome-wide search in yeast cells.<\/p>\n<p>Xiong and PhD\u200d candidate \u200dQinfang Liu discovered that the enzyme ATIC controls the gene at the messenger-RNA \u200dlevel, which instructs the body on how to build proteins. This was \u2063a surprising discovery \u200dfor the team.<\/p>\n<blockquote class=\"wp-block-quote\">\n<p>\u201cThis was a surprising discovery,\u201d Xiong says. \u201cAt the beginning we did a\u2062 screening, we identified a candidate, \u2063and we \u200dfound that it \u2064was targeted at the mRNA level.\u200c This is a new \u2062discovery for us \u200btoo.\u201d<\/p>\n<\/blockquote>\n<p>The researchers then \u2062examined ATIC in human neural cells, as well as fruit\u2063 fly \u200cand mouse models, to understand its impact on Parkinson&#8217;s disease. They \u2062found that AICAr, a \u2064medication that mimics ATIC activity, significantly reduced LRRK2 levels.<\/p>\n<blockquote class=\"wp-block-quote\">\n<p>\u201cWe used a primary neuronal culture to \u200dsee \u200bhow those candidates can regulate LRRK2,\u201d Xiong says. \u201cAnd we found that it can significantly regulate the \u2064LRRK2 expression.\u201d<\/p>\n<\/blockquote>\n<p>AICAr has \u2062shown promise as a therapy for metabolic disorders and cardiovascular illnesses in preclinical studies. However,\u200d its usage \u2063in treating Parkinson&#8217;s disease has been limited \u2064due to the blood-brain\u200c barrier.<\/p>\n<p>To overcome this hurdle, Xiong and her colleagues are\u2064 currently modifying AICAr. They are also collaborating with UConn&#8217;s\u2062 Technology Commercialization Services (TCS)\u2063 to further advance this technology and prepare for clinical human trials.<\/p>\n<p>Stay informed!
